Development of Technologies to Extract and Stabilize DNA from a Broad Spectrum of Microorganisms

We propose to continue with our novel approach to high-efficiency nucleic acids extraction from bacteria. Once complete, DNA Genotek Kits are expected to: be relatively simple to use; be applicable to high-throughput environments; release DNA efficiently from a variety of microorganisms; stabilize the profile of the sample species by (i) stabilizing the released DNA at room temperature for extended periods of time and (ii) preventing overgrowth of microorganisms in collected samples. To assess the efficiency of Kits to extract and stabilize microbial DNA, they will be tested using pure bacteria as well as biological samples containing microorganisms. The ability of the kits to prevent growth/effectively kill bacteria will be tested. The metagenomic DNA of gut microbiota will be analyzed. When Kit development is complete, it is expected to be valuable both for basic researchers and for analyzing clinical samples. The_combination of the Kit (DNA_Genotek)_and the improved_Illumina_sequencing_protocol (University-of-Ottawa) promises_to_improve_the_understanding_of_the_relationship_between_the_microbiota_and_its_effect_on_human_health_and_well-being.
